چکیده
Abstract The maintenance of hemostasis to ensure vascular integrity is dependent upon the rapid conversion zymogen species coagulation cascade their enzymatically active forms. This process culminates in generation serine protease thrombin and polymerization fibrin prevent leak at sites endothelial cell injury or loss cellular junctions. Thrombin can be initiated by extrinsic pathway through exposure blood tissue factor damage, alternatively (F) XII activated foreign surfaces with negative charges, such as glass, contact activation pathway. Here, we used transient particle tracking microrheology investigate mechanical properties response downstream both pathways. We found that structural heterogeneity formation was on reaction kinetics generation. Pharmacological inhibition FXII activity prolonged time form increased degree fibrin, resulting clots reduced properties. Taken together, this study demonstrates a dependency physical biology coagulation.

UNLABELLED BACKGROUND Particle tracking passive microrheology relates recorded trajectories of microbeads, embedded in soft samples, to the local mechanical properties of the sample. The method requires intensive numerical data processing and tools allowing control of the calculation errors. RESULTS We report the development of a software package collecting functions and scripts written in...
متن کاملQuantifying the dynamic transition of hydrogenated castor oil gels measured via multiple particle tracking microrheology.
Rheological modifiers are essential ingredients in commercial materials that exploit facile and repeatable phase transitions. Although rheological modifiers are used to change flow behavior or quiescent stability, the complex properties of particulate gels during dilution is not well studied. We characterize a dynamically evolving colloidal gel, hydrogenated castor oil (HCO), a naturally source...
متن کاملParticle-tracking microrheology of living cells: principles and applications.
A multitude of cellular and subcellular processes depend critically on the mechanical deformability of the cytoplasm. We have recently introduced the method of particle-tracking microrheology, which measures the viscoelastic properties of the cytoplasm locally and with high spatiotemporal resolution. Here we establish the basic principles of particle-tracking microrheology, describing the advan...
متن کاملIntrinsic pathway of coagulation and arterial thrombosis.
Formation of a fibrin clot is mediated by a group of tightly regulated plasma proteases and cofactors. While this system is essential for minimizing blood loss from an injured blood vessel (hemostasis), it also contributes to pathologic fibrin formation and platelet activation that may occlude vessels (thrombosis). Many antithrombotic drugs target key elements of the plasma coagulation mechanis...
متن کاملMicromechanical mapping of live cells by multiple-particle-tracking microrheology.
This paper introduces the method of live-cell multiple-particle-tracking microrheology (MPTM), which quantifies the local mechanical properties of living cells by monitoring the Brownian motion of individual microinjected fluorescent particles. Particle tracking of carboxylated microspheres imbedded in the cytoplasm produce spatial distributions of cytoplasmic compliances and frequency-dependen...
